Supplementary Figures – Alteration of m6A methylation landscape in a mouse model of scleroderma
Supplementary Figure 1. RNA integrity and gDNA contamination test by denaturing agarose gel electrophoresis. The 28S and 18S ribosomal RNA bands should be intense bands. Smaller, more diffuse bands representing low molecular weight RNAs (tRNA and 5S ribosomal RNA) may be present. It is normal to see a diffuse smear of ethidium bromide staining material migrating between the 18S and 28S ribosomal bands, probably comprised of mRNA and other heterogeneous RNA species.
Supplementary Figure 2. Overview of differentially expressed mRNAs in scleroderma. (A) The heat map of expressed mRNAs between the control group and the BLM group. (B) The volcano plot of the expressed mRNAs between the control and BLM groups with Foldchange ≥ 1.5 and p < 0.05.
Supplementary Figure 3. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis of differentially regulated mRNAs. (A) The top ten GO terms of up-regulated mRNAs ranked by Enrichment Score. (B) The top ten GO terms of down-regulated mRNAs ranked by Enrichment Score. (C) The top ten significantly enriched pathways of up-regulated mRNAs ranked by Enrichment Score. (D) The top ten significantly enriched pathways of down-regulated mRNAs ranked by Enrichment Score.
